The son of Theophilus Thornhaugh Gurdon and his wife Anne, of Letton, Norfolk, John Gurdon married Mary Martin Slater Rebow (1805-1842, her second marriage) in 1835. She was the only daughter and heir of Mary Hester Rebow (daughter and co-heir of Isaac Martin Rebow of Wivenhoe Park, Essex) and Gen. Francis Slater Rebow. Like his father-in-law, Gurdon upon his marriage assumed the additional surname of Rebow. After his wife's death in 1842, Gurdon Rebow inherited the Wivenhoe estate. He remarried in 1845 to Lady Georgiana Isabella Toler [d. 1900], daughter of Hector John Graham, 2nd earl of Norbury. They had a son and two daughters; son Hector John Gurdon Rebow inherited the Wivenhoe estate.
